On the physical identity of scintillons: bioluminescent particles in Gonyaulax polyedra.

چکیده
The physical identity of the bioluminescent particles ('scintillons') extracted from the marine dinoflagellate Gonyaulax polyedra was investigated by electron microscopy and particle counts of the active fractions from sucrose density gradients. Membranous structures are abundantly present in purified fractions, while there is a decrease, relative to activity, in the numbers of guanine crystals, which had previously been implicated as bioluminescent structures. The significance of these findings with respect to the in vivo scinrillon is discussed.

The Characterization of Scintillons Bioluminescent particles from the marine dinoflagellate, Gonyaulax polyedra
A new type of biological particle, isolated from the marine dinoflagellate Gonyaulax polyedra, has been partially purified and characterized. When the pH is lowered, the particle emits light in vitro in a fashion closely mimicking the flash of the living cell, and it is referred to as a scintillon (flashing unit). Scintillons are obtained by breaking the cells in buffer at pH 8.2 and purifying ...
متن کاملBioluminescence: mechanism and mode of control of scintillon activity.
Subcellular particles, termed scintillons, isolated from the luminescent dinoflagellate Gonyaulax, emit light when the pH is lowered from 8 to 5.7. The capacity to emit a second flash is regained by incubation of scintillons with the low molecular weight luciferin from Gonyaulax. This compound was previously demonstrated to be the substrate in the bioluminescent reaction catalyzed by a soluble ...
متن کاملThe fine structure of Gonyaulax polyedra, a bioluminescent marine dinoflagellate.
An electron-microscope study of Gonyaulax polyedra, a holophytic marine dinoflagellate which is bioluminescent, has revealed 2 structures not previously described in free-living dinoflagellates: a polyvesicular body occurring at intervals along the cell's periphery, and a large spherical body which may function in intracellular digestion. Coated vesicles were found in the nuclear-Golgi region. ...
متن کاملCharacterization of the bioluminescent organelles in Gonyaulax polyedra (dinoflagellates) after fast-freeze fixation and antiluciferase immunogold staining
To characterize the microsources of bioluminescent activity in the dinoflagellate Gonyaulax polyedra, an immunogold labeling method using a polyclonal antiluciferase was combined with fast-freeze fixation and freeze substitution. The quality of the preservation and the specificity of the labeling were greatly improved compared to earlier results with chemical fixation. Two organelles were speci...
